in this type of environment, on the second router would it be any benefit to set "Forward local domain queries to upstream DNS" for the second router?
 
The only time I can think of where this setting should be used is if your upstream DNS is a Windows server DNS, and that server is authoritative for your LAN domain. This will ensure that your LAN clients will be able to resolve the local domain.

Would that apply to a pi hole also considering that it is a DNS server also.

That statement would still apply. So you would not want forward those queries because the pihole would not be an authoritative DNS server.

Only if that Pi Hole is authoritative for your LAN domain name.

Ahh I understand well I am forcing all my DNS traffic through it so I can filter it, thanks again.
