Is there a way to run a script when DNSFilter/Director settings or iptable rules change? Or maybe just when iptable rules in general change?

If you can make it fall into one of those categories, yes. Otherwise, no. If you're changing the DNSFilter settings via the GUI most likely it is generating a service event so you can use that script. If you're changing them manually you'd need to bounce the service to cause an event most likely.
