First post. I have several home networked computers (wired) which I would like to back up to a USB3 drive.

I can connect the drive to an Asus RT-AC87U or to an i5 computer which is always on and has very little load. Which is better?

Since you have no NAS, I'd put the drive on the i5 always-on computer. Or better, get a big drive inside that i5 computer since that'll be SATA and much faster than USB3. Use network shares.
