I have an RT-AC88U and 4 different laptops with different operating systems. Does it make any difference which laptop is used to upgrade the firmware on the router? It asks for the operating system when selecting the firmware upgrade and I assume they mean on the device being used to upgrade the router firmware.

No, the firmware downloads from Asus' website are OS-independent really. The different Asus utilities are OS-dependent (Device Discovery, Restoration tool, etc.), but rarely needed. As long as you can unzip a ZIP file on your laptop OS, you are good.
