Is it possible to setup a ASUS router with OpenVPN (or any other VPN), there all clients will be behind VPN but one or two clients will be connected to internet directly w/o VPN?
 
Yes, assuming you're using the OpenVPN client on the router, under the Redirect Internet Traffic chose Policy Rules.
