I would say either ext3/ext4 should give you the best performance etc.
Performance wise EXT2 is faster then both EXT3/4 because there is no Journaling.
EXT2 should be used for usb flash drives, they are slow enough as is and EXT3 for hard drives. Lots of people have problems with EXT4, I personally would avoid that unless you require a specific feature which it offers. I am assuming the drives will be dedicated to the router but if you ever want to plug the drives in a Windows computer to transfer stuff and not have to screw around, go with FAT32.
