I have enabled the Enable UPnP Media Server and specified 3 directories that can be accessed. Is there any way to add rules to make this more secure? I've read articles such as this one - https://arstechnica.com/information...e-been-pwned-thanks-to-easily-exploited-flaw/ - that say anyone on the internet can access the files being shared.

I know the article is very old but I could not find anything more recent for ASUS routers in particular. Some still recommend to turn off UPNP altogether.

The Arstechnica article refers to an AiCloud vulnerability that was fixed years ago. Nonetheless, it's not a good idea to deliberately enable remote access to your router if you don't need to.

The UPnP discussion is unrelated to your Media Server.

By remote access, do you mean WAN, telnet & SSH? I have disabled those. I thought the UPnP discussion was related to Media Server because done by UPnP.
