what format for HD?

Okay, I am prepping a WD my pasport HDD 320gb.

1. What format works best with this size HDD on the 68r?

2. Some of you are going to tell me fat***, I remember seeing it some where but I did a search and google search and did not find the specific program that works best to achieve these format types.
